Strain development東京医科歯科大学 平岡優一,田中光一, 2016日本クレア社の C57BL/6JJcl 系統及び DBA2/J 系統を使って得られた BDF1 受精卵の細胞質に Cas9 タンパク質、cr/tracrRNA、ターゲティングプラスミドを顕微注入して作製した。ファウンダー個体を日本クレア社の C57BL/6JJcl 系統に対して 3 世代以上交配してヘテロ型個体を得たのち、同系統との交配にて維持した。へテロ/ホモ型個体ともに生存・繁殖は可能である。
Strain descriptionマウス Actb 遺伝子座に 3’UTR 領域に TRE-FLEX-hM3Dq カセットを有するマウス。カセット内には tetO 配列、minimap CMV promoter 配列、 beta globin intron 配列、loxP 配列、lox2272 配列、i beta ARK 遺伝子、P2A 配列、mCherry 遺伝子、WPRE 配列、human GH polyA 配列が含まれる。tTA・Cre の各分子を発現する別の系統と掛け合わせることで Gq-GPCR 経路を阻害する i beta ARK 遺伝子を複数条件依存的に発現誘導する。
