Strain development | Strain development: The targeting vector harboring neomycin resistance gene with SV40 poly-A addition signal under control of the endogenous UTF1 enhance-promoter was transferred to E14 ES cells to replace the 1st exon of UTF1 gene. Strain description | The prominent phenotype of UTF1 KO homozygous mice is “in utero growth retardation”. This phenotype is due to the growth retardation of placenta, not due to the defect of the differentiation program of embryo per se. This placental growth defect is unexpected because UTF1 gene is identified as a gene whose expression is pluripotent cell specific (Okuda, A. et al. (1998) EMBO J. 17: 2019–2032). This UTF1 KO phenotype indicates that this mouse strain may be useful to analyze not only the differentiation program of the embryo but also the placental developmental program.Homozygous mutant of C57BL/6J background is embryonic or peri-natal lethal. Some of the homozygous mutant mice of F1 (B6 and ICR) genetic background are viable. This phenotypic difference suggests that the severity of phenotype may be dependent on the genetic background (see Nishimoto,M. et al. (2013) PLoS ONE 8(7): e68119).
