Strain development | Developed by Haruhiko Koseki, RIKEN in 2011. M1 ((129 x C57BL/6)F1) ES cells were used. This strain was backcrossed to C57BL/6J. |
Strain description | Tet2 gene floxed mice. Gene targeting for the Tet2 allele was done by flanking exon 9 with Cre-loxP sequences. Conditional deletion of Tet2 (exon 9) can be achieved by crossing the mice with specific cre deleters such as CreERT2. The TET2 protein is a dioxygenase that catalyzes the conversion of 5-methylcytosine (5mC) into 5-hydroxymethylcytosine (5hmC) and further oxidized cytosine (5fC and 5caC), and plays a key role in active DNA demethylation. This conditional allele would be useful to study the roles of Tet2 in DNA de-methylation by combining with tissue specific Cre deleters. |
