Strain development東北大学・大学院・医学系研究科　青木洋子、井上晋一（2014）。BALB/c系統（日本チャルズリバー社）。B6.Cg-Braf〈tm1Tumg〉(RBRC09730)雄マウスとBALB/c雌マウスを10世代戻し交配を行い作製。
Strain descriptionCre存在下でQ241R変異を持つBrafを発現するノックインマウス。BRAFはセリン/スレオニンキナーゼで細胞増殖・分化・アポト―シスに関わるRAS/MAPKシグナル分子のひとつ。BRAFは癌原遺伝子でありその体細胞変異はメラノーマ、甲状腺癌などで多く認められる。対照的にBRAFの生殖細胞系列における変異は心疾患、特異的顔貌、皮膚疾患を特徴とするcardio-facio-cutaneous (CFC)症候群の原因となる。マウスのBraf Q241R変異は、ヒトで最も高頻度に認められるBRAF Q257Rの変異に相当する。CAG-Creマウス (C57BL/6背景) との交配では多くは胎生致死となるが、1-2世代目の解析は可能。Creが存在しなくてもQ241R変異を持つBrafを発現するノックインマウスがRBRC09731として利用可能(ICR背景)。Knock-in mutants in which Braf-mutated protein (BrafQ241R) is expressed in a Cre-dependent manner. Q241R mutation in mouse Braf corresponds to Q257R in human BRAF, the most frequent mutation in the cardio-facio-cutaneous (CFC) syndrome. BrafQ241R heterozygous mice in the ICR genetic background are available as RBRC09731.
