Strain description | ECE-2 (Ece2 / endothelin converting enzyme 2) 遺伝子のノックアウトマウス。エクソン14と15をneoカセットで置換。ホモノックアウトマウスは発達、発育は正常でライフスパンも正常であり、組織学的な異常も認められない。しかしながら、ECE-1ノックアウトマウスと交配させた場合、ECE-1ノックアウトで見られた異常がより重篤になる。また、ECE-1ノックアウトでは見られなかった弁の異常 (abnormal artioventricular valve formation) も観察される。エンドセリン (endothelin、ET) は強力な血管収縮作用を有する血管内皮細胞由来のペプチドで、ブタ大動脈の血管内皮細胞培養上清から、強力な血管収縮作用を有する生理活性物質として単離、精製された。多くの哺乳類には、異なる遺伝子によってコードされるET-1、ET-2、ET-3という3種のペプチド異性体が存在する。また、これらのペプチドの前駆体からの最終的な活性物質エンドセリンの産生に必要なエンドセリン変換酵素 (Endothelin Converting Enzyme-1と-2:ECE-1とECE-2) が同定されている。エンドセリンの受容体にはエンドセリンレセプタータイプAとタイプB (ETAとETB)の2種類が存在する。エンドセリンは心血管系に対する作用のみならず、神経堤由来組織の胚発生において重要な役割を演じていることが示唆されている。 |
