Strain development | EB3/5 ES cells derived from E14tg2aThe homo mice were embryonic lethal. After removing the gene cassette by the cre recombinase, the mice expressed p38alpha Mapk with D316N sevenmaker type mutation and home mice were viable. |
Strain description | The p38alpha (Mapk 14) gene is disrupted by by the insertion of an IRES-β-geo fusion cassette flanked by loxP sites. Homo mice are embryonic lethal.If the cassette is removed by cre mediated recombination, this mouse can conditionally express p38alpha with D316N sevenmaker type mutation (a docking site mutation).A 3.5-kb cassette flanked by two loxP sites is inserted between exons 10 and 11 of p38alpha gene. At the same time exon 11 is replaced by one coding the D316N sevenmaker type mutation. The cassette is generated by modifying the dicistronic targeting construct, pGT1.8IresBgeo (Mountford et al., 1994), consisting of a splice acceptor site (SA) from mouse En-2, an internal ribosomal entry site (IRES) from encephalomyocarditis virus (EMCV), a neomycin phosphotransferase gene (neo), and the SV40 polyadenylation sequence. |
