Strain description | The ERK2 (Mapk 1) gene of ERK2 floxSem mouse is disrupted by by the insertion of an IRES-β-geo fusion cassette flanked by loxP sites. Homo mice are embryonic lethal.If the cassette is removed by cre mediated recombination, this mouse can conditionally express ERK2 with D319N sevenmaker type mutation (a docking site mutation).A 3.5-kb cassette flanked by two loxP sites is inserted after exons 2 of ERK2 gene. At the same time exon 4 is replaced by a partial cDNA of ERK2 starting from exon 3 to the end of the polyadenylation signal sequence. This cDNA codes the D319N sevenmaker type mutation. The cassette is generated by modifying the dicistronic targeting construct, pGT1.8IresBgeo (Mountford et al., 1994), consisting of a splice acceptor site (SA) from mouse En-2, an internal ribosomal entry site (IRES) from encephalomyocarditis virus (EMCV), a neomycin phosphotransferase gene (neo), and the SV40 polyadenylation sequence. |
