Strain descriptionAlcadein beta(Alc beta)は、Alcadein alphaのファミリー分子としてcDNAが単離された神経特異的I型膜タンパク質である((Araki et al [2003] J. Biol.Chem. 278, 49448-49458))。別名calsyntenin3(Clstn3)。AlcbetaはAPP alphaセクレターゼである、ADAM10/ADAM17で切断されAlcbetaCTFを産生する。Alc beta CTFは、さらに gamma セクレターゼで切断され、p3-Alc betaペプチド(主に37アミノ酸からなる)を分泌する(Hata et al [2009] J. Biol. Chem. 284, 36024-36033)。p3-Alcbetaは、人脳脊髄液で検出できる(Hata et al., [2019] Alzheimer & Dement. TRIC in press)。本マウスは、human Alc betaのCTFをPDGF-betaのプロモーター下にTgしたマウスであり、神経細胞でAlc betaCTFを発現し、マウス脳脊髄液中にヒト型p3-Alcbetaを産生する。AlcbetaCTFのアミノ酸配列(human Alc betaのアミノ酸812〜971に相当)のN末にAlc alphaのシグナル配列(アミノ酸1-28)を持つコンストラクトを5‘-PDGF-beta promooter, 3’-SV40 poly (A)を持つDNAをBDF1の受精卵に注入。
