Strain development | Developed by Michael McBurney, Ottawa Regional Cancer Centre and Department of Medicine, University of Ottawa. A knock-in vector carrying a point mutation in exon 5 of Sirt1 gene, in which codon 355 was mutated from CAT (histidine) to TAT (tyrosine) was inserted into exon 5. A hygromycin resistant cassette flanked by loxP sites was removed by crossing with Prm-Cre mice (129-Tg (Prm-Cre) 580g / J). |
